What is Nicotinamide
Nicotinamide is an important compound functioning as a component of the coenzyme NAD. Its primary significance is in the prevention and/or cure of blacktongue and PELLAGRA. Most animals cannot manufacture this compound in amounts sufficient to prevent nutritional deficiency and it therefore must be supplemented through dietary intake. Niacinamide is the active form of vitamin B3 and a component of the coenzyme nicotinamide adenine dinucleotide (NAD). Niacinamide acts as a chemo- and radio-sensitizing agent by enhancing tumor blood flow, thereby reducing tumor hypoxia. This agent also inhibits poly(ADP-ribose) polymerases, enzymes involved in the rejoining of DNA strand breaks induced by radiation or chemotherapy.
How Nicotinamide Works
It is believed that sulbutiamine works similarly to thiamine however the effects are greatly enhanced. Thiamine is essential in the body and even more essential for cognitive function because it is used in the biosynthesis of acetylcholine and GABA.  sulbutiamine is believed to have effects on the cholinergic, glutamatergic and dopamine systems which result in its nootropic benefits.
Thiamine is inferior to sulbutimaine in its absorption rate. It is absorbed through the intestines at a poor rate that is often reduced by hindrances like folic deficiency. Having higher concentrations of Thiamine in the digestive tract will also reduce this absorption rate even further. Sulbutiamine is much more lipophilic and is able to dissolve in fats. This not only gives it a better absorption rate but more importantly, increases blood brain barrier efficiency.
Health Benefits of Nicotinamide
To date, research on the potential health benefits of Nicotinamide is somewhat limited. However, there's some evidence that Nicotinamide may help with certain health conditions. Here's a look at some key study findings:
Diabetes
Nicotinamide may help prevent insulin-dependent diabetes mellitus, also known as type 1 diabetes. Type 1 diabetes is an autoimmune disease that results when the insulin-producing beta-cells of the pancreas get destroyed by your body's own white blood cells. According to a study published in the "The British Journal of Clinical Practice" in 1992, nicotinamide may protect beta-cells from damage and improve insulin secretion in high-risk patients. However, more research is needed to evaluate the effects of nicotinamide in treatment of type 1 diabetes.
Cancer
Nicotinamide may also play a role in preventing cancer. According to the Linus Pauling Institute, NAD can influence cellular response to DNA damage and help prevent cancer development. Taking increased doses of niacin with antioxidants decreased esophageal, throat and mouth cancers by 40 percent. However, more research is needed to evaluate whether taking nicotinamide has the same effects.
Pellagra
Taking nicotinamide can help prevent a condition called pellagra, caused by vitamin B3 deficiency. Pellagra can cause dark red skin rash on your hands, feet, calves, neck and face, gastrointestinal problems, fatigue, insomnia and memory loss. If left untreated, pellagra can be fatal. Pellagra is treated by taking high doses of nicotinamide or niacin.
Nicotinamide Side Effects
High doses of niacinamide cause skin flushing due to the relaxation of blood vessels near the surface of the skin. Also, taking niacinamide in prescription-strength dose can lead to the breakdown of muscle tissue and heart palpitations.
Nicotinamide Dosage
The recommended federal daily intake of niacinamide is 20 mg. Prescription doses of niacinamide range as high as 2,500 mg per day, but this should only be done under a doctor's supervision.
